چکیده

16 wHat’s On tHe weB Cytogenetically normal acute myeloid leukemia (CyN-AmL) makes up 40 to 50 percent of cases of adult AmL. The heterogeneous nature of CyN-AmL makes prognosis uncertain, and despite the clinical heterogeneity, induction therapy remains largely uniform. Technological advances in DNA sequencing, however, are providing new insights into the molecular basis of CyN-AmL, suggesting new diagnostic and prognostic categories and novel approaches to therapy.

Mutations in epigenetic modifiers in the pathogenesis and therapy of acute myeloid leukemia.

Recent studies of the spectrum of somatic genetic alterations in acute myeloid leukemia (AML) have identified frequent somatic mutations in genes that encode proteins important in the epigenetic regulation of gene transcription. This includes proteins involved in the modification of DNA cytosine residues and enzymes which catalyze posttranslational modifications of histones. Here we describe th...

Covalent Modification of Bacteriophage T4 DNA Inhibits CRISPR-Cas9

UNLABELLED The genomic DNAs of tailed bacteriophages are commonly modified by the attachment of chemical groups. Some forms of DNA modification are known to protect phage DNA from cleavage by restriction enzymes, but others are of unknown function. Recently, the CRISPR-Cas nuclease complexes were shown to mediate bacterial adaptive immunity by RNA-guided target recognition, raising the question...

Metabolic Alterations in Cancer Cells and the Emerging Role of Oncometabolites as Drivers of Neoplastic Change

The mitochondrion is an important organelle and provides energy for a plethora of intracellular reactions. Metabolic dysregulation has dire consequences for the cell, and alteration in metabolism has been identified in multiple disease states-cancer being one. Otto Warburg demonstrated that cancer cells, in the presence of oxygen, undergo glycolysis by reprogramming their metabolism-termed "aer...

Review Series EPIGENETICS IN HEMATOLOGY Mutations in epigenetic modifiers in the pathogenesis and therapy of acute myeloid leukemia

Acute myeloid leukemia (AML) is a clonal disorder of hematopoiesis characterized by the accumulation of immature myeloid cells accompanied by impaired normal hematopoiesis. Given the need to improve outcome in AML, multiple studies aimed at genetic characterization of AML have been performed in the hopes of furthering our understanding of AML pathogenesis and identifying new therapeutic approac...

DNA methylation regulates neuronal glutamatergic synaptic scaling.

Enhanced receptiveness at all synapses on a neuron that receive glutamatergic input is called cell-wide synaptic upscaling. We hypothesize that this type of synaptic plasticity may be critical for long-term memory storage within cortical circuits, a process that may also depend on epigenetic mechanisms, such as covalent chemical modification of DNA. We found that DNA cytosine demethylation medi...
